Expression of Lactate Dehydrogenase A-Gene in Regenerating Rat Liver

Abstract
We have investigated the induction of rat liver LDH A-gene during the pre-replicative phase of regenerating rat liver. In vitro run-off transcriptional assay showed that rat liver LDH A-gene transcription activity was sharply increased at 14 hours after partial hepatectomy (PH). And the accumulation of LDH A-mRNA became apparent at 15 hours and progressively increased to peak at 24 hours after PH. The western blot analysis also showed that the apparent increase of LDH A-isozyme level in liver cytoplasmic fraction was detectable at 15 hours and reached plateau after 21 hours and kept its level until 24 hours after PH. The increased level of nuclear LDH A-isozyme was also detectable at 21 hours and progressively increase up to 24 hours after PH. The temporal increase of LDH A-gene expression of rat liver was shown to coincide well with the initiation of DNA replication in regenerating rat hepatocytes. It was also shown that the accumulations of LDH A-mRNA in regenerating liver (the concentrations of LDH A-isozyme) were totally abolished after injection of (D,L)-propranolol. With the above results in mind, we hypothesize that the induction mechanism of LDH A-gene in regenerating rat liver may be mediated by beta-adrenergic receptor. And also we propose that the newly synthesized LDH A-subunit may be translocated into the nucleus and plays a role in the initiation of DNA replication after partial hepatectomy.
